    Production of extracellular protease from bacterial co-culture using solid state fermentation
    (UMT Lahore, 2019) Qurat-ul-ain
    Proteases are enzymes that hydrolyze proteins into amino acids by cleaving peptide bonds. They are also known as peptidase or proteinase. Out of total 82% commercially reported enzymes, 60% of them are hydrolytic enzymes to which protease accounts for 20% of total usage. Various sources can be utilized to obtain proteases such as animal, bacteria, fungi and plants. Besides other sources, microbial protease are preferred because of the presence of all desired characteristics for industrial applications. This study aims to isolate and screen high potential protease producing bacterial strains from soil samples and to produce protease from bacterial co-culture using solid state fermentation. The bacterial strains were isolated from soil samples using a specified media. The strains were then sub-cultured and preserved on Nutrient agar media. The preserved strains were tested for their compatibility and then were further screened using SSF. After fermentation, enzyme assay was carried out. Then finally, the strains of co-culture 19 that showed the highest absorbance were further identified using morphological and biochemical tests. It was found that the strain 1 of co-culture 19 was observed as cocci, irregular and it was positive for gram staining, endospore staining, casein hydrolysis test, catalase test and methyl red test. As for strain 2, it was identified as bacillus rod-shaped and was positive for gram staining, casein hydrolysis test, catalase test and methyl red test but negative for endospore staining. From this study, it was concluded that, the strains of co-culture 19 can be an important co-culture for enzyme production in soil state fermentation by utilizing wheat bran as substrate. These results need to confirmed at large scale for statistical analysis.
