Item Synthesis of copper oxide nanoparticles conjugated with methotrexate and estimation of targeted drug delivery(UMT, Lhr, 2022) Abida Nisar KhattakCancer is one of the most prevalent cause of death while the conventional methods for its treatment have numerous negative effects that make nanotechnology necessary for therapeutics utilized for cancer treatment. Methotrexate (MTX), is a potent anticancer medication used to treat different malignancies. In current study the Copper oxide nanoparticles were prepared by using chemical method and conjugated with methotrexate. The gluteraldehyde method was performed to bind the CuO NPs with methotrexate with 5% concentration of gluteraldehyde. The nanoparticles were observed under microscope for cross check. The characterization of CuO NPs as well as their conjugation with drug were done by light microscope, UV-Visible spectrophotometry, and FT-IR. Spectrophotometric analysis of CuO NPs was carried out at different wavelengths to check absorption spectra. FT-IR was performed to observe binding of functional groups. Anticancer properties were checked on MCF-7 cells through MTT assay and immunohistochemistry. MTT assay was performed to observe cell death rate against MCF-7 cells while immunohistochemistry was performed for checking the attachment of composite drug with mouse epithelial cancerous tissues. The results showed appearance of brown clumps that confirms the formation of CuO NPs under light microscope. The absorbance shown at 320nm by using UV-Visible spectrophotometry shows the conjugation of CuO NPs with cysteine while 330nm shows the conjugation of CuO NPs with drug. In FT-IR analysis the peaks between 1500-1600 range is high that indicates the conjugation of MTX with CuO NPs. MTX conjugated CuO NPs showed positive results by inhibiting proliferation of MCF-7 cells upon the concentration of 500ng/mL through MTT assay while positive attachment of composite to clump of mouse epithelial cells was shown through immunohistochemistry. The results showed that CuO conjugated with methotrexate have enhanced anticancer activity and show high cytotoxicity in MCF-7 cells and it can also be used in medicines to treat cancer.
